How can we understand Paul's knowledge in the mysteries of Christ?

Paul's grasp of the mysteries of Christ came by direct revelation from the Lord Jesus Himself, not from men. The mystery was God’s secret, held in His mind until He revealed it through Paul’s epistles. When believers read those epistles, they may understand those mysteries of Christ.

Les teaches that Paul’s knowledge was not humanly acquired; it came "by revelation" when the Lord Jesus Himself made known unto him the mystery. This mystery is the composite of truth for the church age, and it comes from Paul’s pen. In Ephesians 3:4, Paul says that when we read his epistles, we may understand his knowledge in the mysteries of Christ. Verse 5 explains that these things were not made known to the sons of men as they are now revealed, showing God’s pattern of keeping a secret until He is ready to reveal it, and then expecting mankind to believe it. The fellowship of the mystery was hid in the mind of God from the beginning of the world, and Paul was given grace to preach among the Gentiles the unsearchable riches of Christ, which are beyond human comprehension.
